Biological Background: Siglec-8 Function and Localization

  • Siglec-8 (Sialic acid-binding Ig-like lectin 8), also known as SAF-2 (sialoadhesin family member 2), is a transmembrane receptor belonging to the Siglec family of sialic acid-binding lectins encoded by the SIGLEC8 gene (GeneID: 27181).
  • Functions as a putative adhesion molecule mediating sialic-acid dependent binding to blood cells. Preferentially binds to α-2,3-linked sialic acid and, to a lesser extent, α-2,6-linked sialic acid. The binding site may be masked by cis interactions with sialic acids on the same cell surface. Related references: PMID:10625619 PMID:10856141
  • Recognizes glycan epitopes containing a terminal N-acetylneuraminic acid (sialic acid) and an underlying 6-O-sulfated galactose, with a strong preference for 6-O-sulfated sialyl-Lewis X structures. Related references: PMID:27357658
  • The protein features multiple immunoglobulin-like domains, a transmembrane helix, and a cytoplasmic tail. Alternative splicing generates isoforms with predicted molecular weights of 44, 46, and 54 kDa, consistent with its mature glycosylated form.
  • Localized to the plasma membrane, Siglec-8 is specifically expressed on blood cells, particularly basophils, mast cells, and eosinophils, making it a key marker for these lineages.
  • Post-translational modifications include N-glycosylation and disulfide bond formation, essential for structural stability and ligand recognition. The three-dimensional structure has been experimentally determined (PDB available), highlighting its lectin domain architecture.

Experimental Guidance and Technical Tips

  • The immunogen covers the extracellular sialic acid-binding domain (aa 17–363), so this antibody is likely to recognize native conformations – consider using it primarily for surface staining in flow cytometry.
  • Validated for flow cytometry (FC) with human samples; for other applications (e.g., Western blot, IHC), in-house optimization including antigen retrieval and blocking conditions is advised.
  • Due to restricted expression on eosinophils, mast cells, and basophils, include appropriate positive controls (e.g., purified eosinophils or cell lines) and isotype controls to confirm specificity.
  • Titrate the antibody for optimal signal-to-noise ratio, as Siglec-8 expression levels may vary among cell types.
