Biological Background: ROR1 Function and Localization

  • ROR1 is an inactive tyrosine-protein kinase transmembrane receptor, encoded by the ROR1 gene (also known as NTRKR1). It belongs to the receptor tyrosine kinase family and features immunoglobulin-like and kringle domains.
  • Alternative splicing produces a short isoform that is highly expressed in fetal and adult central nervous system (CNS) and various cancers, whereas the full-length isoform is expressed in heart, lung, and kidney.
  • ROR1 has very low kinase activity in vitro and is unlikely to function as a tyrosine kinase in vivo. Related references: PMID:25029443
  • It acts as a receptor for WNT5A, activating the NFkB signaling pathway and potentially inhibiting WNT3A-mediated signaling. Related references: PMID:25029443 PMID:27162350
  • In the inner ear, ROR1 is crucial for spiral ganglion neurons to innervate auditory hair cells; mutations in ROR1 are associated with non-syndromic deafness. Related references: PMID:27162350
  • Through its ligand IGFBP5, ROR1 forms a complex with ERBB2 to enhance CREB oncogenic signaling, highlighting its role in cancer. Related references: PMID:36949068
  • Subcellularly, ROR1 localizes to the cell membrane and is found on axonal projections, consistent with its role in cell signaling and neuronal development.
  • Post-translational modifications include N-linked glycosylation, and the protein contains disulfide bonds critical for its domain architecture.

Experimental Guidance and Technical Tips

  • This antibody was raised against the extracellular domain (amino acids 24-403), making it suitable for detecting cell surface ROR1 in live-cell flow cytometry.
  • It is directly conjugated to , a bright and photostable fluorophore, simplifying experimental workflows by eliminating secondary staining.
  • For FC, titrate the antibody to determine the optimal concentration, and include appropriate isotype controls and viability dyes for accurate gating.
  • Validation is recommended in relevant human cell lines or primary samples known to express ROR1, such as certain cancer cell lines or neuronal cells.
