Biological Background: ROR1 Function and Localization
- ROR1, also known as Neurotrophic tyrosine kinase receptor-related 1 (NTRKR1), is an inactive tyrosine-protein kinase transmembrane receptor that serves as a receptor for WNT5A. It has very low kinase activity and is unlikely to function as a tyrosine kinase in vivo. Related references: PMID:25029443
- Binding of WNT5A to ROR1 activates the NFkB signaling pathway and may inhibit WNT3A-mediated signaling, implicating it in non-canonical Wnt pathways. Related references: PMID:25029443, PMID:27162350
- In the inner ear, ROR1 is crucial for spiral ganglion neurons to innervate auditory hair cells, highlighting its developmental role. Related references: PMID:27162350
- Through interaction with IGFBP5, ROR1 forms a complex with ERBB2 to enhance CREB oncogenic signaling, contributing to cancer progression. Related references: PMID:36949068
- ROR1 is localized to the cell membrane and cell projections such as axons, consistent with its role in cell surface signaling.
- Alternative splicing produces a short isoform that is strongly expressed in fetal and adult CNS and in various human cancers, while the full-length protein is prominently expressed in heart, lung, and kidney.
- ROR1 contains immunoglobulin-like and kringle domains, and is modified by glycosylation and disulfide bonds, which may influence its stability and ligand interactions.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 24-403) encompasses the extracellular region of human ROR1; consider this design when planning epitope competition or domain-specific studies.
- This antibody is validated for flow cytometry (FC). Optimize the antibody concentration by titration on appropriate positive and negative control cells to achieve optimal staining.
- Because ROR1 is a cell surface receptor, the conjugate may be suitable for live-cell staining without permeabilization; however, confirm in your specific experimental system.
- For other applications or species cross-reactivity, validate the antibody under your own conditions, as the provided data only cover human reactivity.
