Biological Background: Podoplanin Function and Localization

  • Podoplanin (PDPN), also known as Aggrus, Gp36, or T1-alpha, is a type I transmembrane glycoprotein with multiple isoforms generated by alternative splicing.
  • It mediates cell migration and adhesion by interacting with various partners: binding to CLEC1B triggers platelet activation/aggregation, while CD9 attenuates this response. Related references: PMID:14522983, PMID:15231832, PMID:17222411, PMID:18541721
  • PDPN promotes epithelial-mesenchymal transition (EMT) via MSN or EZR, leading to RHOA activation and increased cell migration and invasiveness. Related references: PMID:17046996, PMID:21376833
  • Interaction with CD44 drives directional cell migration in epithelial and tumor cells, while binding to LGALS8 may link the lymphatic endothelium to the extracellular matrix. Related references: PMID:20962267, PMID:19268462
  • In tumor cells, PDPN controls invadopodia stability and extracellular matrix degradation via RHOC-ROCK-LIMK-CFL1 signaling. Related references: PMID:25486435
  • Subcellular localization includes membrane raft, lamellipodium, filopodium, microvillus, ruffle, invadopodium, and apical/basolateral membranes; also detected in the cytosol.
  • Highly expressed in placenta, lung, skeletal muscle, and brain; weakly in kidney and liver. Up-regulated in colorectal tumors and a subset of oral squamous cell carcinomas.
  • Heavily glycosylated with sialic acid modifications, contributing to the observed multiple molecular weight forms (12–24 kDa).

Experimental Guidance and Technical Tips

  • The immunogen spans the extracellular domain (aa 21–123), making this antibody suitable for detection of surface Podoplanin by flow cytometry.
  • For FC, titrate the antibody to determine the optimal working dilution using a known Podoplanin-expressing human cell line (e.g., lymphatic endothelial cells or tumor cell lines).
  • Include appropriate isotype controls and viability dyes to ensure specific staining and exclude dead cells.
  • Given the multiple glycosylation forms, confirm expression patterns in the relevant cell system; the observed signal intensity may vary with glycosylation state.
  • This antibody is validated for human reactivity; cross-reactivity with other species is not expected.
