Biological Background: PLZF Function and Localization
- Zinc finger and BTB domain-containing protein 16 (ZBTB16), also known as promyelocytic leukemia zinc finger protein (PLZF) or zinc finger protein 145 (ZNF145), is a transcription factor characterized by multiple zinc finger motifs.
- Functions as a transcriptional repressor by recruiting histone deacetylases to target gene promoters (PubMed:10688654, PubMed:24359566).
Related references: PMID:10688654 PMID:24359566 - Probable substrate-recognition component of an E3 ubiquitin-protein ligase complex, mediating ubiquitination and proteasomal degradation of target proteins (PubMed:14528312).
Related references: PMID:14528312 - Plays a role in myeloid maturation and the development or maintenance of differentiated tissues.
- Localizes to the nucleus, notably in nuclear bodies, consistent with its role in transcriptional regulation.
- Expressed in hematopoietic tissues including bone marrow and early myeloid cells, and also found in ovary, kidney, and lung.
- Undergoes phosphorylation and is involved in chromosomal rearrangements associated with acute promyelocytic leukemia; recognized as a proto-oncogene.
Experimental Guidance and Technical Tips
- For intracellular flow cytometry, follow standard fixation and permeabilization protocols appropriate for nuclear antigens.
- The antibody was generated against a recombinant protein corresponding to amino acids 6-137 of human PLZF, an N-terminal region; epitope mapping may be useful for specific applications.
- Due to human cross-reactivity, validate the antibody in species-specific contexts if non-human samples are to be analyzed.
- Titration experiments are recommended to determine the optimal antibody concentration for your specific cell type and experimental conditions.
- Ensure appropriate isotype controls are used for accurate gating in flow cytometry.
