Biological Background: PLZF Function and Localization

  • Zinc finger and BTB domain-containing protein 16 (PLZF), also known as ZNF145, is encoded by the ZBTB16 gene. It belongs to the BTB/POZ domain-containing zinc finger transcription factor family.
  • Acts as a transcriptional repressor, mediated at least in part through the recruitment of histone deacetylases to target promoters. Related references: PMID:10688654, PMID:24359566
  • Plays a role in myeloid maturation and the development and/or maintenance of other differentiated tissues.
  • Functions as a probable substrate-recognition component of an E3 ubiquitin-protein ligase complex, mediating ubiquitination and subsequent proteasomal degradation of target proteins. Related references: PMID:14528312
  • Localizes to the nucleus, including nuclear bodies, consistent with its function in transcriptional regulation.
  • Within the hematopoietic system, PLZF is expressed in bone marrow, early myeloid cell lines, and peripheral blood mononuclear cells. It is also found in the ovary, and at lower levels in the kidney and lung.
  • Contains nine C2H2-type zinc fingers and a BTB/POZ domain; known to undergo alternative splicing and is associated with chromosomal rearrangements in acute promyelocytic leukemia.
