Biological Background: PI3K Regulatory Subunits (p85α/p85β/p55γ) Function and Localization
- The phosphatidylinositol 3-kinase (PI3K) regulatory subunit p85α (encoded by PIK3R1) is an adapter protein that plays a critical role in mediating the activation of the PI3K/AKT signaling pathway. It contains SH2 and SH3 domains that bind to phosphorylated tyrosine residues on activated receptor tyrosine kinases and adaptor proteins, recruiting the p110 catalytic subunit to the plasma membrane to produce PIP3 and propagate downstream signals. - This antibody is designed to recognize the phosphorylated forms of three PI3K regulatory subunits: p85α (PIK3R1), p85β (PIK3R2), and p55γ (PIK3R3), specifically at the conserved tyrosine phosphorylation sites Y467 (p85α), Y464 (p85β), and Y199 (p55γ). These phosphorylation events are associated with activation of PI3K signaling downstream of growth factor receptors.
- p85α is necessary for insulin-stimulated glucose uptake and glycogen synthesis in insulin-sensitive tissues and plays a role in signaling in response to various growth factor receptors (e.g., FGFR, KIT, PDGFR). It also modulates the cellular response to ER stress by promoting nuclear translocation of XBP1 isoform 2, thereby contributing to glucose tolerance improvement. Related references: PMID:20348923
- Subcellularly, p85α is predominantly localized to the cytoplasm, where it interacts with membrane-bound receptors upon stimulation.
- Isoform 2 of p85α shows tissue-specific expression, being present in skeletal muscle and brain, and at lower levels in kidney and cardiac muscle. At the protein level, isoforms 2 and 4 are detected in skeletal muscle.
