Biological Background: Perforin-1 Function and Localization
- Perforin-1 (PRF1, also known as cytolysin or lymphocyte pore-forming protein PFP) is a 61 kDa pore-forming glycoprotein that plays a key role in granzyme-mediated programmed cell death and immune defense against virus-infected and neoplastic cells. Related references: PMID:20889983, PMID:21037563, PMID:24558045
- It is essential for killing non-self target cells, such as in transplant rejection and certain autoimmune diseases. Related references: PMID:9058810
- In its calcium-bound form, Perforin inserts into the target cell membrane, oligomerizes, and forms large transmembrane pores, causing cytolysis and facilitating apoptosis. Related references: PMID:20889983, PMID:21037563
- Perforin mediates the uptake of cytotoxic granzymes into target cells, promoting efficient delivery of cationic cargo, while neutral or anionic proteins are transferred poorly. Related references: PMID:20038786, PMID:20225066, PMID:24558045
- Subcellularly, Perforin localizes to cytolytic granules, is secreted, and can be found in the endosome lumen and on the cell membrane. It contains EGF-like domains, disulfide bonds, and is a transmembrane beta-strand protein.
- Clinically, mutations in PRF1 are associated with familial hemophagocytic lymphohistiocytosis (FHL), a severe hyperinflammatory disorder.
Experimental Guidance and Technical Tips
- The immunogen corresponds to nearly full-length mature Perforin (aa 21–555); the resulting antibody recognizes native and intracellular forms.
- conjugation enables direct detection by intracellular flow cytometry without a secondary reagent; we recommend optimizing fixation and permeabilization protocols for your specific cell types.
- The predicted molecular weight is 61 kDa; however, glycosylation and other modifications may result in additional bands or shifts in certain sample systems—validate in the relevant biological context.
- For best performance, titrate the antibody in your assay to determine the optimal working concentration.
