Biological Background: P4HB Function and Localization
- P4HB, also known as protein disulfide-isomerase, is a multifunctional enzyme that catalyzes the formation, breakage, and rearrangement of disulfide bonds in nascent proteins.
- It acts as a reductase at the cell surface, cleaving disulfide bonds of exofacial proteins and thereby modulating cell surface protein structure.
- At high concentrations, following phosphorylation by FAM20C, P4HB functions as a chaperone that inhibits aggregation of misfolded proteins. Related references: PMID:32149426
- At low concentrations, it can promote aggregation (anti-chaperone activity), and it also serves as a structural subunit of prolyl 4-hydroxylase and microsomal triacylglycerol transfer protein.
- P4HB is a receptor for LGALS9 on Th2 T helper cells, enhancing cell migration by increasing plasma membrane disulfide reductase activity. Related references: PMID:21670307
- Subcellularly, it resides primarily in the endoplasmic reticulum lumen but is also found in melanosomes and at the cell membrane.
