Biological Background: P4HB Function and Localization

  • P4HB, also known as protein disulfide-isomerase, is a multifunctional enzyme that catalyzes the formation, breakage, and rearrangement of disulfide bonds in nascent proteins.
  • It acts as a reductase at the cell surface, cleaving disulfide bonds of exofacial proteins and thereby modulating cell surface protein structure.
  • At high concentrations, following phosphorylation by FAM20C, P4HB functions as a chaperone that inhibits aggregation of misfolded proteins. Related references: PMID:32149426
  • At low concentrations, it can promote aggregation (anti-chaperone activity), and it also serves as a structural subunit of prolyl 4-hydroxylase and microsomal triacylglycerol transfer protein.
  • P4HB is a receptor for LGALS9 on Th2 T helper cells, enhancing cell migration by increasing plasma membrane disulfide reductase activity. Related references: PMID:21670307
  • Subcellularly, it resides primarily in the endoplasmic reticulum lumen but is also found in melanosomes and at the cell membrane.
  • Post-translational modifications include acetylation and phosphorylation; it contains a redox-active CXXC motif and multiple thioredoxin domains.
  • Mutations in P4HB are associated with human disorders such as craniosynostosis and osteogenesis imperfecta.

Experimental Guidance and Technical Tips

  • The immunogen targets amino acids 329–508, covering the C-terminal region. This design may detect full-length P4HB and potential splice variants containing this region.
  • For intracellular flow cytometry, use optimized fixation/permeabilization reagents to ensure epitope accessibility, as P4HB is primarily located in the ER.
  • Confirm expression in your target cell type; P4HB is widely expressed but levels may vary.
  • If adapting to other applications (e.g., IHC, WB), validate antibody specificity under the respective experimental conditions.
