- Chondroitin sulfate proteoglycan 4 (CSPG4), also known as NG2, MCSP, MCSPG, MSK16, CSPG4A, HMW-MAA, or MEL-CSPG, is a transmembrane proteoglycan expressed on various cell types.
- It plays a key role in cell proliferation and migration, stimulating endothelial cell motility during microvascular morphogenesis. NG2 may also inhibit neurite outgrowth and growth cone collapse during axon regeneration.
- The extracellular domain serves as a cell surface receptor for collagen alpha-2(VI) and binds growth factors and extracellular matrix proteases, modulating their activity and contributing to tissue remodeling.
- Through its cytoplasmic C-terminus, NG2 functions as a signal transducer, recruiting scaffolding and signaling proteins to promote retraction fiber formation, cell polarization via Rho GTPase activation, and integrin-mediated adhesion through CDC42, ACK1, and BCAR1 cascades.
