Biological Background: MMP9 Function and Localization
- Matrix metalloproteinase-9 (MMP9), also known as gelatinase B, 92 kDa gelatinase, or 92 kDa type IV collagenase, is a member of the zinc-metalloproteinase family involved in extracellular matrix degradation.
- Plays an essential role in local proteolysis of the extracellular matrix and in leukocyte migration. Related references: PMID:12879005, PMID:1480034, PMID:2551898
- Cleaves KiSS1 at a Gly-|-Leu bond and NINJ1 to generate the Secreted ninjurin-1 form. Related references: PMID:12879005, PMID:32883094
- Degrades type IV and type V collagen into large C-terminal fragments and shorter N-terminal fragments, and also degrades fibronectin. Related references: PMID:1480034
- Secreted into the extracellular space and localizes to the extracellular matrix.
- Detected in neutrophils, normal alveolar macrophages, and granulocytes. Related references: PMID:7683678
- Contains calcium-binding and zinc-binding sites, disulfide bonds, and is initially produced as a zymogen that requires activation.
Experimental Guidance and Technical Tips
- This antibody is directly conjugated with , making it suitable for intracellular flow cytometry (FC intra) without a secondary reagent.
- For intracellular staining, a suitable permeabilization and fixation protocol is necessary; consider titrating the antibody to determine optimal working concentration in the relevant cell system.
- MMP9 is secreted; thus, pre-treatment with a protein transport inhibitor (e.g., brefeldin A) may be required to retain the protein in appropriate intracellular compartments before staining.
- Validate the staining specificity using known MMP9-positive controls, such as stimulated neutrophils or macrophage cell lines, and appropriate isotype controls.
