Biological Background: LILRA2/CD85h Function and Localization
- LILRA2 is a type I transmembrane receptor belonging to the leukocyte immunoglobulin-like receptor (LILR) family; it is also referred to as CD85h, immunoglobulin-like transcript 1 (ILT-1), or leukocyte immunoglobulin-like receptor 7 (LIR-7).
- It functions as a pattern recognition receptor in innate immunity, specifically binding to N-terminally truncated immunoglobulin molecules generated by bacterial and fungal proteases from pathogens such as Legionella pneumophila, Mycoplasma hyorhinis, Streptococcus pneumoniae, Staphylococcus aureus, and Candida albicans.

- For flow cytometry, consider using appropriate isotype controls and titrating the antibody to achieve optimal staining on primary human innate immune cells (monocytes, neutrophils) or LILRA2-transfected cell lines.
- In immunofluorescence/ICC, since LILRA2 is a cell surface receptor, gentle fixation (e.g., 4% PFA) without permeabilization may help preserve surface staining; if intracellular staining is needed, permeabilization may be adjusted.
