Biological Background: LAMP2 Function and Localization
- LAMP2 (Lysosome-associated membrane glycoprotein 2), also known as CD107b, LAMP-2, LGP-96, or LGP110, is a heavily glycosylated transmembrane protein encoded by the LAMP2 gene. Through alternative splicing, it produces three major isoforms: LAMP-2A, LAMP-2B, and LAMP-2C, which differ in their cytosolic tails and exhibit distinct tissue distributions.
- As a key lysosomal membrane glycoprotein, LAMP2 plays an essential role in lysosome biogenesis and pH regulation by acting as a direct inhibitor of the proton channel TMEM175, thereby facilitating lysosomal acidification and optimal hydrolase activity. Related references: PMID:37390818
- LAMP2 is a central mediator of chaperone-mediated autophagy (CMA), a selective degradation pathway. It binds cytosolic chaperone complexes (e.g., HSPA8/HSC70) that deliver target proteins such as GAPDH, GPX4, NLRP3, and MLLT11 to the lysosomal membrane, where LAMP2 acts as a receptor to translocate substrates into the lysosome for degradation. Related references: PMID:11082038, PMID:18644871, PMID:24880125, PMID:36586411
- LAMP2 is required for the fusion of autophagosomes with lysosomes during macroautophagy; cells lacking LAMP2 fail to accumulate STX17 on autophagosomes, which prevents fusion and impairs degradation of autophagic cargo. Related references: PMID:27628032
- LAMP2 facilitates efficient MHC class II-mediated presentation of exogenous antigens by enabling lysosomal degradation and subsequent loading of antigenic peptides onto nascent MHC II molecules, though it is dispensable for endogenous antigen presentation. Related references: PMID:15894275, PMID:20518820
- Subcellularly, LAMP2 localizes primarily to the lysosomal membrane, but is also found in endosomal, autophagosomal, and plasma membranes, reflecting its dynamic trafficking and functions in endolysosomal and autophagic pathways.
- Isoform-specific expression patterns are observed: LAMP-2A is highly expressed in placenta, lung, and liver, while LAMP-2B and LAMP-2C show distinct distributions across tissues such as spleen, thymus, brain, and small intestine. Related references: PMID:26856698, PMID:7488019
- Notably, LAMP2 is associated with Danon disease, a glycogen storage disorder, and its study involves keywords such as alternative splicing, autophagy, disulfide bond, glycoprotein, and transmembrane helix.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 29-375 of human LAMP2, covering a large luminal domain common to all isoforms; this broad coverage supports detection of multiple LAMP2 variants where sequence homology permits.
- The antibody is directly conjugated to , enabling one-step staining for immunofluorescence (IF/ICC) and intracellular flow cytometry (FC intra) without the need for secondary antibodies.
- For intracellular flow cytometry, fixation and permeabilization are required; use standard protocols (e.g., 4% PFA fixation followed by saponin or Triton X-100 permeabilization) and optimize incubation times and antibody concentrations for your specific cell type.
- Validate antibody performance in the relevant biological system (human) and include appropriate positive and negative controls (e.g., wild-type versus LAMP2-knockdown cells) to confirm specificity.
- Since LAMP2 is heavily glycosylated and exists in multiple isoforms, apparent molecular weight by Western blot may differ from the predicted 44-45 kDa; testing by WB is not listed, so if needed, consider pilot experiments to verify detection capability.
