Biological Background: LAMP2 Function and Localization
- LAMP2 (Lysosome-associated membrane glycoprotein 2), also known as CD107b, LGP-96, and LAMPB, is encoded by the LAMP2 gene (GeneID: 3920) and exists as multiple splice isoforms (LAMP-2A, LAMP-2B, LAMP-2C) with distinct expression profiles.
- Plays an important role in lysosome biogenesis, lysosomal pH regulation and autophagy. Related references: PMID:11082038, PMID:18644871, PMID:24880125
- Acts as a direct inhibitor of the proton channel TMEM175, facilitating lysosomal acidification for optimal hydrolase activity. Related references: PMID:37390818
- Functions as a key receptor in chaperone-mediated autophagy (CMA), binding target proteins (e.g., GAPDH, GPX4, NLRP3, MLLT11) and delivering them to lysosomes for degradation. Related references: PMID:11082038, PMID:18644871, PMID:24880125, PMID:36586411
- Required for the fusion of autophagosomes with lysosomes by promoting accumulation of STX17 on autophagosomes, enabling normal degradation of autophagic contents. Related references: PMID:27628032
- Contributes to efficient MHC class II-mediated presentation of exogenous antigens via its role in lysosomal protein degradation. Related references: PMID:15894275, PMID:20518820
- Localizes primarily to the membranes of lysosomes, endosomes, autophagosomes, and the cell membrane.
- Isoform LAMP-2A is highly expressed in placenta, lung, and liver; LAMP-2B is widely expressed; and LAMP-2C is detected in intestine, colon, heart, brain, and skeletal muscle.
Experimental Guidance and Technical Tips
- The immunogen spans the luminal domain (aa 29-375) of human LAMP2, allowing recognition of all three major isoforms; confirm specificity in the relevant sample system.
- For immunofluorescence (IF/ICC), use appropriate permeabilization to access the lysosomal/endosomal membrane pool; co-staining with lysosomal markers (e.g., LAMP1, cathepsins) may aid localization.
- For intracellular flow cytometry (FC intra), optimize fixation and permeabilization conditions to preserve epitope; include isotype controls and consider pre-incubation with Fc block to minimize non-specific binding.
- The dye is photostable and suitable for multiplex IF or multi-color flow cytometry; ensure instrument compensation settings are optimized.
