Biological Background: LAMP1/CD107a Function and Localization
- LAMP1/CD107a, encoded by the LAMP1 gene, is a heavily glycosylated lysosomal membrane protein also known as LGP120 and CD107a.
- It plays a central role in lysosome biogenesis and pH regulation by inhibiting the proton channel TMEM175, thereby facilitating lysosomal acidification necessary for hydrolase activity (PubMed:37390818). Related references: PMID:37390818
- In NK cells, LAMP1/CD107a is critical for cytotoxicity by mediating granule movement and perforin trafficking to lytic granules, and it protects NK cells from self-damage by their own cytolytic contents (PubMed:2022921, 23632890, 23847195). Related references: PMID:2022921 PMID:23632890 PMID:23847195
- Presents carbohydrate ligands to selectins, contributing to cell adhesion (PubMed:7685349). Related references: PMID:7685349
- Acts as a receptor for Lassa virus, promoting viral entry and fusion, and supports FURIN-mediated cleavage of mumps virus fusion protein (PubMed:24970085, 25972533, 27605678, 29295909). Related references: PMID:24970085 PMID:25972533 PMID:27605678 PMID:29295909
- Subcellularly, LAMP1/CD107a localizes to lysosomes, late endosomes, endosomes, cell membrane, and cytolytic granule membranes.
- Post-translational modifications include extensive N-linked glycosylation, disulfide bond formation, and alternative splicing.
Experimental Guidance and Technical Tips
- Since this antibody targets an intracellular epitope (immunogen: amino acids 29-382), it is intended for intracellular flow cytometry. Optimize permeabilization and fixation protocols using appropriate controls.
- is a bright far-red fluorophore; ensure proper compensation when multiplexing with other dyes and use an unlabeled control to check for non-specific binding.
- Validate antibody staining in human cell lines expressing LAMP1/CD107a; typical lysosomal staining patterns can be confirmed by co-localization with lysosomal markers.
- For degranulation assays (e.g., NK cell activation), this intracellular antibody requires permeabilization, so it may detect total cellular protein. Adjust gating strategies accordingly.
- Store at recommended conditions and protect from light to preserve fluorophore stability.
