Biological Background: LAMP1/CD107a Function and Localization
- LAMP1, also known as CD107a or LGP120, is a heavily glycosylated lysosomal membrane protein involved in lysosome biogenesis and homeostasis.
- It regulates lysosomal pH by directly inhibiting the TMEM175 proton channel, facilitating acidification for optimal hydrolase activity. Related references: PMID:37390818
- LAMP1 plays a critical role in NK cell cytotoxicity: it mediates cytotoxic granule movement, perforin trafficking, and protects NK cells from self-inflicted damage. Related references: PMID:2022921, PMID:23632890, PMID:23847195
- The protein presents carbohydrate ligands to selectins, supporting cell adhesion. Related references: PMID:7685349
- LAMP1 acts as a host cell receptor for Lassa virus glycoprotein and promotes virus-host membrane fusion. Related references: PMID:24970085, PMID:25972533, PMID:27605678, PMID:29295909
- It also supports FURIN-mediated cleavage of mumps virus fusion protein F by interacting with both FURIN and unprocessed viral F protein.
- Subcellularly, LAMP1 localizes to lysosome, late endosome, endosome, cell membrane, and cytolytic granule membranes.
- The mature glycoprotein migrates at approximately 38–45 kDa in SDS-PAGE due to heavy glycosylation.
Experimental Guidance and Technical Tips
- The immunogen (aa 29-382) covers the luminal domain of LAMP1, suggesting the antibody recognizes an epitope in this region.
- For intracellular flow cytometry (FC intra), optimize fixation and permeabilization conditions to enable antibody access to lysosomal compartments.
- Include appropriate isotype controls and validate staining in relevant human cell lines or primary samples.
- LAMP1 can transiently appear on the cell surface; thus, evaluate both surface and intracellular staining protocols as needed.
- Due to glycosylation heterogeneity, the observed molecular weight in western blot may range from 38 to 45 kDa; confirm band specificity if adapting to other applications.
