Biological Background: KLRG1 Function and Localization
- KLRG1 (Killer cell lectin-like receptor subfamily G member 1) is a type II transmembrane glycoprotein belonging to the C-type lectin superfamily; it is also known as MAFA, CLEC15A, or MAFA-L.
- The protein functions as an inhibitory receptor on natural killer (NK) cells and T-cells, primarily CD8+ T-cells, and negatively regulates their effector functions upon ligand engagement.
- KLRG1 recognizes classical cadherins such as E-cadherin (CDH1), N-cadherin (CDH2), and R-cadherin (CDH4), and this interaction is thought to mediate "missing self" recognition, sensing the integrity of cadherin-expressing tissues.
- The receptor contains an immunoreceptor tyrosine-based inhibitory motif (ITIM) in its cytoplasmic domain, which mediates inhibitory signaling.
- KLRG1 is expressed specifically on NK cells and T-cells, with predominant expression on CD8+ T-cell subsets; it is often used as a marker of terminally differentiated or senescent T-cells.
- Subcellularly, KLRG1 localizes to the cell membrane via a signal-anchor transmembrane domain.
- Post-translational modifications include N-linked glycosylation and disulfide bond formation, which are characteristic of its extracellular C-type lectin domain. Alternative splicing has been reported.
