Biological Background: KIR2DL4/CD158d Function and Localization
- KIR2DL4 (killer cell immunoglobulin-like receptor 2DL4), also known as CD158d, G9P, and KIR103AS, is a member of the immunoglobulin superfamily and functions as a receptor for non-classical MHC class Ib molecule HLA-G. It recognizes HLA-G in complex with beta-2 microglobulin (B2M) and a nonamer self-peptide. In decidual NK cells, binding to the peptide-bound HLA-G-B2M complex triggers NK cell senescence-associated secretory phenotype, promoting vascular remodeling and fetal growth during early pregnancy. Related references: PMID:16366734, PMID:23184984, PMID:29262349
- This receptor may play a critical role in balancing immune tolerance and antiviral immunity at the maternal-fetal interface by regulating the effector functions of NK cells, CD8+ T cells, and B cells. Related references: PMID:10190900, PMID:16366734
- Upon ligand engagement, KIR2DL4 initiates signaling from the endosomal compartment, leading to activation of PRKDC-XRCC5 and AKT1, culminating in an NF-kappa-B-dependent pro-inflammatory response. Related reference: PMID:20179272
- Subcellularly, KIR2DL4 is localized to the cell membrane and early endosome membrane, where it can initiate signaling upon endocytosis.
- KIR2DL4 is predominantly expressed in decidual NK cells and innate lymphoid cell type I (ILC1), with a subset of peripheral NK cells also positive. Related references: PMID:29262349, PMID:19304799
- The protein undergoes alternative splicing and glycosylation, yielding multiple isoforms with molecular weights ranging from approximately 24 kDa to 41 kDa, consistent with the protein weight pattern observed for this antibody.
Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 24-242 of human KIR2DL4, corresponding to the extracellular domain, ensuring recognition of the native receptor.
- As an -conjugated rabbit monoclonal antibody, it is ready for direct flow cytometry without the need for secondary detection reagents.
- Optimize antibody titration for best signal-to-noise ratio in your specific experimental system; typical starting dilutions can be determined empirically.
- Since KIR2DL4 is expressed on a subset of NK cells, consider enriching for CD56+ NK cells or using appropriate cell lines (e.g., NKL, NK-92) for validation.
- Multiple protein isoforms may be detected by Western blot due to alternative splicing and glycosylation, but this product is validated for flow cytometry.
