Biological Background: JAM-A/CD321/F11R Function and Localization

  • JAM-A, encoded by the F11R gene, is a single-pass type I membrane protein and member of the immunoglobulin superfamily (IgSF). It is also known as junctional adhesion molecule 1 (JAM-1), platelet F11 receptor, and platelet adhesion molecule 1 (PAM-1).
  • Plays a critical role in epithelial tight junction assembly, recruiting the PARD3 polarity protein to early cell junctions. Related references: PMID:11489913
  • Functions as a ligand for integrin αL/β2 (LFA-1), facilitating memory T-cell and neutrophil transmigration across endothelial and epithelial barriers. Related references: PMID:11812992
  • Involved in platelet activation; engagement of JAM-A on platelets contributes to thrombus formation and hemostasis. Related references: PMID:10753840
  • Serves as a cellular receptor for certain viruses, including Mammalian reovirus sigma-1 and Human Rotavirus strain Wa, mediating viral entry.
  • Localizes to tight junctions of polarized epithelial and endothelial cells, as well as the cell membrane of circulating leukocytes and platelets.
  • Expressed in endothelium, epithelium, and leukocytes at the protein level.
  • Post-translational modifications include N-linked glycosylation and phosphorylation; contains a signal peptide, two Ig-like C2-type domains, and a transmembrane helix.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the extracellular domain (aa 28-242) of human JAM-A, covering both Ig-like domains; this region is well-suited for detection of surface-expressed JAM-A in live-cell flow cytometry.
  • For flow cytometry, consider optimizing antibody titration and include appropriate isotype controls to assess non-specific binding.
  • JAM-A is expressed on endothelial cells, epithelial cells, and leukocytes; validate expression in your target sample using positive (e.g., activated leukocytes) and negative controls.
  • The conjugate is compatible with standard flow cytometers equipped with a red laser (~647 nm); protect from light during staining and handling.
  • For adherent cells, use gentle dissociation methods to preserve surface epitopes.
  • Cross-reactivity is reported for human; to use with other species, verify sequence homology and perform empirical validation.
