Biological Background: IL9 Function and Localization

  • Interleukin-9 (IL9), also known as Cytokine P40 or T-cell growth factor P40, is a multifunctional cytokine primarily secreted by T-helper 2 lymphocytes, mast cells, and NKT cells.
  • It plays important roles in the immune response against parasites and affects intestinal epithelial permeability and adaptive immunity. Related references: PMID:29742432
  • IL9 induces the differentiation of specific T-cell subsets, including IL-17-producing helper T-cells (TH17), and promotes the proliferation and differentiation of mast cells.
  • The biological effects of IL9 are mediated through a receptor complex consisting of the IL9R subunit and the signal-transducing IL2RG subunit, leading to the rapid activation of JAK1 and JAK3 kinases and subsequent STAT1, STAT3, and STAT5-mediated transcriptional programs.
  • STAT1 alone mediates the induction of differentiation genes, while STAT3 and STAT5 are involved in protecting cells from apoptosis.
  • IL9 is a secreted glycoprotein featuring a signal peptide, pyrrolidone carboxylic acid modification, and a known 3D structure.
  • The calculated molecular weight of the unprocessed precursor is approximately 15.9 kDa, consistent with the observed 15 kDa mature form.

Experimental Guidance and Technical Tips

  • For intracellular flow cytometry detection, optimize permeabilization and fixation protocols to detect IL9 in the appropriate cell type, such as activated T-cells or mast cells.
  • In immunofluorescence/immunocytochemistry applications, verify antibody specificity using IL9-positive cell lines or tissue samples and appropriate negative controls.
  • Because the immunogen spans the mature cytokine region, the antibody may recognize both precursor and fully processed forms; validate performance in the relevant sample system.
  • Consider co-staining with lineage markers to identify IL9-producing cell subsets in mixed populations.
