Biological Background: IL-6R/CD126 Function and Localization

• Interleukin-6 receptor subunit alpha (IL-6R, CD126/gp80) is a transmembrane glycoprotein that exists as a membrane-bound form and a soluble isoform (sIL6R) generated by alternative splicing and proteolytic shedding.

• Membrane-bound IL-6R binds interleukin-6 (IL-6) with low affinity; signal transduction requires association with the gp130 co-receptor to activate JAK/STAT, MAPK, and other pathways, regulating immune responses, acute-phase reactions, and hematopoiesis.
