Biological Background: Interleukin-5 (IL-5) Function and Localization

  • Interleukin-5 (IL-5), also known as eosinophil differentiation factor (EDF) and T-cell replacing factor (TRF), is a cytokine belonging to the IL-5 family. It is a secreted glycoprotein with a molecular weight of approximately 15 kDa.
  • IL-5 promotes the survival, differentiation, and chemotaxis of eosinophils, playing a central role in eosinophil-mediated immune responses. Related references: PMID:2653458 PMID:9010276
  • IL-5 also acts on B-cells, inducing immunoglobulin production, growth, and differentiation (By similarity).
  • IL-5 signals through a heterodimeric receptor composed of the IL-5 receptor alpha chain (IL5RA) and the common beta chain (CSF2RB). This triggers activation of kinases such as LYN, SYK, and JAK2, leading to downstream RAS-MAPK and JAK-STAT5 pathways. Related references: PMID:1495999 PMID:22528658 PMID:7613138
  • IL-5 is predominantly produced by T-lymphocytes and NK cells, and is present in peripheral blood mononuclear cells.
  • The protein is secreted and contains structural features such as disulfide bonds and multiple glycosylation sites. Alternative splicing of the IL5 gene results in different isoforms.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the mature IL-5 protein (amino acids 20-134), ensuring recognition of the native secreted form.
  • For intracellular flow cytometry, appropriate fixation and permeabilization protocols are required. Consider titrating the antibody to optimize staining index.
  • Validate performance in relevant sample types, such as activated T-cell cultures or PBMCs, and include appropriate controls.
