Biological Background: IFN-gamma Function and Localization
- Interferon gamma (IFN-γ), also known as immune interferon, is a type II interferon.
- Produced primarily by activated T lymphocytes and NK cells, and secreted to exert its effects.
- Binds to its receptor IFNGR1 and signals through the JAK-STAT pathway, leading to STAT1 activation and transcription of IFN-γ-regulated genes. Related references: PMID:16914093 PMID:8349687
- Plays a key role in antimicrobial, antiviral, and antitumor immunity by activating effector immune cells and enhancing antigen presentation. Related references: PMID:16914093 PMID:8666937
- Enhances MHC class I antigen presentation by inducing immunoproteasome subunit replacement and PA28 expression, improving peptide generation and repertoire. Related references: PMID:8666937 PMID:8163024 PMID:11112687
- Up-regulates MHC class II complexes by promoting expression of cathepsins B, H, and L. Related references: PMID:7729559
- Participates in hematopoietic stem cell regulation, affecting their development, quiescence, and differentiation (By similarity).
- Post-translational modifications include glycosylation, signal peptide cleavage, and formation of pyrrolidone carboxylic acid.
Experimental Guidance and Technical Tips
- The immunogen encompasses amino acids 24–166 of human IFN-γ, corresponding to the mature secreted form; thus the antibody is expected to recognize the biologically active cytokine.
- For intracellular flow cytometry (FC intra), optimize fixation and permeabilization protocols and include appropriate activation controls (e.g., PMA/ionomycin-stimulated T cells).
- Consider titrating the antibody to determine the optimal staining concentration for your specific sample type.
- Validate performance in the relevant cellular context (e.g., activated T lymphocytes or NK cells) using known positive and negative controls.
