Biological Background: Interferon alpha/beta receptor 2 Function and Localization

  • IFNAR2 (interferon alpha/beta receptor 2), encoded by the IFNAR2 gene (aliases: IFNABR, IFNARB), is a subunit of the type I interferon receptor. Multiple splice variants yield proteins with molecular weights of approximately 58 kDa, 37 kDa, and 27 kDa.

  • Activated JAKs phosphorylate specific tyrosine residues on the intracellular tails of IFNAR1 and IFNAR2, creating recruitment sites for STAT1 and STAT2.   • A naturally occurring soluble isoform of IFNAR2 (isoform 3) is secreted and acts as a potent inhibitor of type I IFN signaling, serving as a regulatory decoy receptor.
  • The receptor is predominantly localized to the cell membrane but is also found in secreted form; it is widely expressed in blood cells and certain tumor cell lines, consistent with its role in innate antiviral immunity.

  • This antibody was generated using a recombinant protein immunogen corresponding to amino acids 28-237 of human IFNAR2. Researchers may consider using a similar immunogen strategy for generating antibodies against the extracellular domain.
  • The antibody is validated for flow cytometry (FC) with human samples. For other applications, we recommend performing pilot experiments to determine optimal working concentrations and fixation/permeabilization conditions.
  • Given the existence of multiple splice isoforms, confirm the recognized isoform(s) by western blot or other assays if needed, and consider the potential presence of soluble IFNAR2 in sample supernatants.
