Biological Background: IFN-alpha/beta R2 Function and Localization
- IFNAR2 encodes the interferon alpha/beta receptor 2, which together with IFNAR1 forms the heterodimeric receptor for type I interferons (IFN-α, -β, -ε, -ω, -κ). Type I interferon binding activates the JAK-STAT signaling cascade, leading to transcriptional regulation of interferon-responsive genes.
Related references: PMID:10049744, PMID:10556041, PMID:21854986 - Mechanistically, ligand binding brings IFNAR1 and IFNAR2 into close proximity, driving cross-phosphorylation of associated Janus kinases (JAK1 on IFNAR2, TYK2 on IFNAR1). This phosphorylation creates docking sites for STAT transcription factors (STAT1, STAT2), which are then phosphorylated, dimerize, and translocate to the nucleus to modulate gene expression.
Related references: PMID:11682488, PMID:12105218, PMID:28165510, PMID:9121453 - IFNAR2 also functions as a potent inhibitor of type I interferon receptor activity. A soluble secreted isoform (isoform 3) is detected in urine and may modulate systemic interferon signaling.
Related references: PMID:7759950, PMID:8181059 - The protein localizes to the cell membrane and is also secreted. It is expressed in blood cells, lymphoblastoid, and fibrosarcoma cell lines.
- Key structural and post-translational features include alternative splicing, disulfide bonds, glycosylation, and phosphorylation, giving rise to multiple isoforms with molecular weights of 58 kDa, 37 kDa, and 27 kDa.
- The gene is also known by aliases IFNABR, IFNARB, and is critical for innate antiviral immune responses.
Experimental Guidance and Technical Tips
- For flow cytometry, titrate the antibody to determine the optimal working concentration for your specific cell types; human PBMCs or IFNAR2-expressing cell lines can serve as positive controls.
- Include appropriate isotype controls and fluorescence-minus-one (FMO) controls to accurately assess background and antigen-specific staining.
- This antibody may detect multiple IFNAR2 isoforms. Validate detection in your sample system and consider complementary methods (e.g., Western blot) to confirm molecular weight bands.
- Store upon receipt at 2–8°C, protected from light, to maintain the fluorophore stability.
