Biological Background: ICAM2/CD102 Function and Localization
- Intercellular adhesion molecule 2 (ICAM2, CD102) is a cell surface glycoprotein belonging to the immunoglobulin superfamily, functioning as a ligand for integrin LFA-1 (ITGAL:ITGB2) and dendritic cell receptor DC-SIGN (CD209).
- ICAM2 mediates leukocyte cell-cell adhesion on resting cells, ensuring critical immune functions such as antigen presentation, lymphocyte recirculation, and immune surveillance. Related references: PMID:10077629, PMID:1676048, PMID:2497351
- Through interaction with DC-SIGN, ICAM2 triggers transendothelial migration of dendritic cell precursors, facilitating immune cell trafficking from blood into peripheral and lymphoid tissues. Related references: PMID:11017109
- Subcellularly, ICAM2 localizes to the cell membrane and is enriched on microvilli, consistent with its role in cellular projections for adhesion.
- Constitutively expressed on leukocytes, including resting lymphocytes and monocytes, but absent in neutrophils; also highly expressed on vascular endothelial cells and lymphatic vessels in tonsils. Related references: PMID:1676048, PMID:11017109
- Structurally, ICAM2 contains immunoglobulin-like domains, disulfide bonds, and a transmembrane region; its mature protein has a predicted molecular weight of 30.6 kDa (approximately 31 kDa observed).
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (aa 25-223), ensuring the antibody can recognize surface-expressed ICAM2 on live cells in flow cytometry applications.
- As an conjugate, this antibody is ready-to-use for direct flow cytometry without secondary detection; consider titrating to optimize the signal-to-noise ratio for your specific cell system.
- This antibody is specific for human ICAM2; cross-reactivity with other species has not been reported, so validate in the relevant sample system if non-human models are used.
- For intracellular staining protocols requiring permeabilization, confirm that the epitope remains accessible under the chosen fixation and permeabilization conditions.
