Biological Background: HER2/ErbB2 Function and Localization
- Receptor tyrosine-protein kinase erbB-2 (HER2/ErbB2), also known as proto-oncogene Neu, p185erbB2, or CD340, is a member of the epidermal growth factor receptor family.
- Functions as an essential component of neuregulin-receptor complexes; requires coreceptors for ligand binding and does not interact with neuregulins alone. GP30 is a potential ligand.
- Regulates outgrowth and stabilization of peripheral microtubules via the MEMO1-RHOA-DIAPH1 signaling pathway, leading to GSK3B inhibition and subsequent membrane localization of APC and CLASP2 for microtubule capture.
- In the nucleus, acts as a transcriptional regulator: binds to the PTGS2/COX-2 promoter (5'-TCAAATTC-3') and activates its transcription; also implicated in CDKN1A transcriptional activation involving STAT3 and SRC.
- Activates rRNA gene transcription by RNA polymerase I, enhancing protein synthesis and cell growth.
- Subcellular localization: primarily at the cell membrane, including ruffle membranes; also detected in early endosomes, cytoplasm (perinuclear), and nucleus.
- Post-translational modifications include phosphorylation (phosphoprotein), glycosylation (glycoprotein), and disulfide bond formation; the protein is a transmembrane tyrosine kinase with ATP-binding activity.
- Expressed in various tumor tissues including primary breast tumors and tumors from small bowel, esophagus, kidney, and mouth.
