Biological Background: HER2/ErbB2 Function and Localization

  • Receptor tyrosine kinase that is a member of the epidermal growth factor receptor (EGFR/ErbB) family and functions as an essential component of neuregulin-receptor complexes, although neuregulins do not directly bind to it. It requires a coreceptor for ligand binding.
  • Upon activation, mediates downstream signaling including the MEMO1-RHOA-DIAPH1 pathway that inhibits GSK3B, leading to stabilization of microtubules at the cell membrane via APC, CLASP2, and MACF1, thereby regulating cell morphology and motility.
  • Also localizes to the nucleus where it acts as a transcription factor, binding the PTGS2/COX-2 promoter at the 5'-TCAAATTC-3' sequence to activate transcription, and co-activates CDKN1A expression with STAT3 and SRC. It also enhances rRNA synthesis and protein translation.
  • Subcellular localization includes the cell membrane, ruffles, early endosomes, cytoplasm (perinuclear region), and nucleus, reflecting its dynamic signaling and transcriptional roles.
  • Post-translational modifications include autophosphorylation at tyrosine residues upon ligand stimulation, and the protein is a glycoprotein with disulfide bonds.
  • Overexpressed in a variety of tumor tissues including primary breast tumors and tumors from small bowel, esophagus, kidney and mouth, and is a well-known oncogene frequently amplified or mutated in cancer.
