Biological Background: Glypican 3 Function and Localization
- Glypican-3 (GPC3) is a cell surface proteoglycan attached to the membrane via a GPI-anchor, belonging to the glypican family of heparan sulfate proteoglycans. Related references: PMID:14610063
- Negatively regulates the hedgehog signaling pathway by competing with the receptor PTC1 for hedgehog protein binding, leading to internalization and lysosomal degradation of the complex.
- Positively regulates the canonical Wnt signaling pathway by binding to the Wnt receptor Frizzled and promoting Frizzled-Wnt ligand interaction. Related references: PMID:16227623 PMID:24496449
- Inhibits the dipeptidyl peptidase activity of DPP4, modulating substrate cleavage and downstream cellular processes. Related references: PMID:17549790
- Plays a role in limb patterning, skeletal development, and renal branching morphogenesis by modulating growth factor responses (BMP2, BMP7, FGF7).
- Localized to the cell membrane; detected in placenta, lung, liver, and kidney.
- Contains heparan sulfate chains, a GPI-anchor, and undergoes alternative splicing; classified as a glycoprotein, lipoprotein, and phosphoprotein with disease variant associations.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 481–580, near the C-terminal region of human GPC3; confirm that the antibody recognizes the intended form (full-length or shed) in your system.
- This conjugate is suitable for direct flow cytometry; titrate the antibody to determine the optimal working dilution for your cell type.
- Validate reactivity using GPC3-positive cell lines (e.g., HepG2, A549) or primary tissue samples known to express GPC3.
- Because GPC3 is GPI-anchored, a standard surface staining protocol without permeabilization may be employed.
