Biological Background: FATP1 (SLC27A1) Function and Localization
- FATP1, also known as solute carrier family 27 member 1 (SLC27A1), is a long-chain fatty acid transport protein that facilitates the import of long-chain fatty acids (LCFA) at the plasma membrane (PubMed:12556534, PubMed:20530735, PubMed:21395585). Related references: PMID:12556534 PMID:20530735 PMID:21395585
- It also functions as an acyl-CoA ligase (very long-chain acyl-CoA synthetase), catalyzing ATP-dependent formation of fatty acyl-CoA from LCFA and very-long-chain fatty acids (VLCFA), thereby trapping fatty acids intracellularly and promoting further uptake.
- The protein localizes to the cell membrane, endomembrane system, and cytoplasm, consistent with its dual role in transport and metabolic activation of fatty acids.
- Highest expression is observed in muscle and adipose tissue, with moderate levels in small intestine, and low to undetectable levels in liver (PubMed:10873384, PubMed:21395585). It is also expressed in brain gray matter (PubMed:21395585). Related references: PMID:10873384 PMID:21395585
- FATP1 likely participates in fatty acid transport across the blood-brain barrier (PubMed:21395585) and may influence cholesterol metabolism by similarity. Related references: PMID:21395585
- Alternative splicing generates multiple isoforms; the UniProt canonical sequence has a predicted molecular weight of 71.1 kDa, while the product datasheet notes detection of bands at 52 kDa and 71 kDa, possibly reflecting isoforms or post-translational modifications.
- Key biological processes associated with FATP1 include fatty acid metabolism, lipid transport, and long-chain fatty acid import.
Experimental Guidance and Technical Tips
- This antibody is supplied as an conjugate, allowing direct detection in intracellular flow cytometry without the need for a secondary antibody.
- The immunogen corresponds to amino acids 510–646 of human FATP1 (C-terminal region), which is conserved across isoforms; however, detection of specific isoforms should be verified in the relevant cell type.
- For intracellular staining, optimize cell fixation and permeabilization protocols; methanol or saponin-based permeabilization may be suitable for detecting membrane-associated proteins.
- Titrate the antibody to determine the optimal concentration for your specific assay system, as performance can vary with cell type and target abundance.
- Include appropriate negative controls (e.g., isotype control) and confirm compensation settings for the fluorophore.
- Due to the hydrophobic nature of FATP1, ensure that sample preparation does not lead to protein aggregation or loss.
