Biological Background: EGFR Function and Localization

• EGFR (epidermal growth factor receptor) is a member of the ErbB family of receptor tyrosine kinases and is also known as ErbB1 or HER1.
• It binds EGF family ligands, including EGF, TGF-alpha, amphiregulin (AREG), epigen (EPGN), betacellulin (BTC), epiregulin (EREG), and heparin-binding EGF-like growth factor (HBEGF), leading to receptor homo- or heterodimerization and autophosphorylation on key cytoplasmic residues. Experimental Guidance and Technical Tips

• This antibody targets an extracellular domain epitope (amino acids 335-525), making it suitable for detection of cell surface EGFR in live-cell and fixed-cell applications like flow cytometry and immunofluorescence.
• For flow cytometry, we recommend titrating the antibody to determine the optimal concentration and including appropriate isotype controls to ensure specificity.
• For immunofluorescence, gentle fixation with paraformaldehyde followed by permeabilization with Triton X-100 or methanol may be used; however, detection of surface EGFR may not require permeabilization.
• Since cross-reactivity has been validated only for human EGFR, users should verify reactivity in other species or sample types of interest.
• As with any recombinant antibody, perform batch-to-batch consistency checks and validate in the intended assay system.
