Biological Background: Cytokeratin 8 Function and Localization
- Cytokeratin 8 (CK-8, K8, KRT8) is a type II keratin intermediate filament protein encoded by the KRT8 gene.
- Together with its obligate type I partner KRT18, it forms the keratin intermediate filament network that provides mechanical stability and resilience to simple epithelial cells. Related references: PMID:26823019
- It protects cells from non-mechanical stress, such as apoptosis (By similarity).
- Required for ARHGEF40-mediated actin stress fiber formation and tensional force-induced stress fiber formation and reinforcement. Related references: PMID:26823019
- In striated muscle, together with KRT19, it may contribute to linking the contractile apparatus to dystrophin at the costameres. Related references: PMID:16000376
- Subcellularly, it localizes to the cytoplasm and cytoskeleton, and is also found in the nucleus, nucleoplasm, and nuclear matrix.
- Tissue expression includes simple epithelia; observed in muscle fibers at costameres, and in gingival mucosa and hard palate of the oral cavity.
- Post-translational modifications include acetylation, phosphorylation, methylation, and ubiquitination; it is also a glycoprotein with potential isopeptide bonds, reflecting its dynamic regulation.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 1-483 of human Cytokeratin 8, providing a broad antigenic region; however, as a monoclonal antibody, it recognizes a single epitope within this sequence.
- Validated for intracellular flow cytometry (FC intra); optimizing fixation and permeabilization steps is recommended, with methanol permeabilization often suitable for cytoskeletal targets.
- The predicted molecular weight is 53-56 kDa; if applied in Western blotting, consider validating under reducing conditions to account for potential protein complexes.
- Cross-reactivity is confirmed for human; for other species, assess sequence homology to determine suitability.
