Biological Background: Cytokeratin 19 Function and Localization
- Cytokeratin 19 (CK-19), also known as Keratin-19 (K19) and K1CS, is a type I intermediate filament protein encoded by the KRT19 gene.
- It plays a structural role in myofibers by linking the contractile apparatus to dystrophin at costameres, in coordination with KRT8.
- Expressed in a broad range of epithelial tissues: basal keratinocytes of hair follicles, sweat gland, mammary gland, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium.
- Also found in vascular smooth muscle cells, umbilical cord vascular walls, and striated muscle fibers where it localizes to costameres.
- Contains a central coiled-coil rod domain typical of keratins, facilitating intermediate filament assembly.
- Subject to post-translational phosphorylation and methylation, which may regulate filament dynamics.
- Has been identified in host-virus interaction pathways and is listed as a reference proteome member with direct protein sequencing evidence.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a peptide within amino acids 301-400 of human Cytokeratin 19 (NP_002267.2). Consider verifying linear epitope accessibility in your target cell type.
- This antibody is directly conjugated to , making it suitable for intracellular flow cytometry. Use appropriate permeabilization and fixation protocols as required for nuclear/cytoplasmic antigens.
- Cross-reactivity is reported for human. If using with other species, validate specificity beforehand.
- Include relevant isotype controls and single-stain compensation controls in your flow cytometry panel design.
