Biological Background: Cytokeratin 19 Function and Localization
- Cytokeratin 19 (KRT19) is a type I keratin intermediate filament protein encoded by the KRT19 gene. It is also known as CK-19, K19, and K1CS.
- Together with its type II partner keratin 8 (KRT8), it forms heterodimers that assemble into intermediate filaments, providing structural support to cells.
- In striated muscle, KRT19 links the contractile apparatus to dystrophin at costameres, contributing to myofiber organization and sarcolemma stability.
- KRT19 is predominantly expressed in simple and glandular epithelia, including sweat glands, mammary ducts, bile ducts, gastrointestinal tract, bladder urothelium, and ectocervical epithelium. It is also found in basal keratinocytes of hair follicles and a subset of vascular wall cells.
- This keratin is widely used as a marker for epithelial cells and in cancer research to detect circulating tumor cells and characterize carcinomas.
- Post-translational modifications include phosphorylation and methylation, which may regulate filament dynamics and protein-protein interactions.
- The protein contains coiled-coil domains essential for dimerization and filament assembly.
Experimental Guidance and Technical Tips
- This antibody is designed for intracellular flow cytometry (FC). Ensure effective permeabilization and fixation protocols for optimal intracellular staining.
- The immunogen is a synthetic peptide corresponding to amino acids 301–400 of human KRT19, which lies within a cytoplasmic region. Validate reactivity in your specific sample type and experimental system.
- For multiplex flow cytometry, confirm spectral compatibility of the conjugate with other fluorophores and instrument configurations.
- Include appropriate negative controls (e.g., KRT19-negative cell lines or isotype controls) and positive controls (e.g., epithelial cell lines) to assess staining specificity.
- Titrate the antibody concentration for your particular assay; a starting range of 1–5 µg/mL is often used in flow cytometry, but optimal dilutions must be determined empirically.
