Biological Background: Cytokeratin 18 Function and Localization
- Keratin 18 (CK-18, K18) is a type I intermediate filament protein and the major keratin partner of type II keratin 8 (KRT8) in simple epithelial cells. It provides mechanical stability and resilience to epithelial tissues. Related references: PMID:15529338, PMID:26823019
- KRT8/KRT18 filaments mediate ARHGEF40-dependent actin stress fiber formation and reinforcement in response to tensional forces, and act downstream of ROCK kinase in a mechanical stress feedback loop. Related references: PMID:26823019
- The organized orientation of KRT8/KRT18 filaments contributes to the elongated morphology of epithelial tubules.
- Phosphorylation of KRT18 is critical for filament reorganization and dynamics. Related references: PMID:7523419, PMID:8522591, PMID:9298992
- KRT18 participates in the delivery of mutated CFTR to the plasma membrane and in interleukin-6 (IL-6)-mediated barrier protection. Related references: PMID:15529338, PMID:16424149, PMID:17213200
- It is involved in the hepatic uptake of thrombin-antithrombin complexes.
- Subcellularly, keratin 18 localizes to the nucleus matrix, nucleolus, perinuclear cytoplasm, and cytoplasmic intermediate filament networks.
- Tissue expression is prominent in colon, placenta, and liver, with weak expression in exocervix; expression increases in lymph nodes of breast carcinoma.
Experimental Guidance and Technical Tips
- Consider using this -conjugated antibody for intracellular flow cytometry detection of cytokeratin 18 in permeabilized human epithelial cells or carcinoma cell lines.
- Optimize fixation and permeabilization conditions for your specific sample type to maintain epitope recognition.
- Validate the antibody in your experimental system, as CK-18 expression levels can vary with cell differentiation and tissue origin.
- When multiplexing, ensure spectral compatibility of with other fluorophores in your panel.
