Biological Background: CLEC12A Function and Localization
- CLEC12A (CD371, CLL-1, MICL) is a single-pass type II transmembrane receptor of the C-type lectin family. Alternative splicing produces multiple isoforms.
- Functions as a myeloid inhibitory receptor; upon ligand engagement, its ITIM motif is phosphorylated, recruiting tyrosine-protein phosphatases PTPN6 and PTPN11 to dampen SYK-mediated activation signals, thereby suppressing myeloid cell responses including cytokine production and degranulation. - Acts as a pattern-recognition receptor for diverse ligands: recognizes DNA in neutrophil extracellular traps (NETs), monosodium urate crystals, and pathogen-associated molecules such as plasmodium hemozoin and mycobacterial mycolic acid, linking it to anti-inflammatory feedback and host defense. - The immunogen used corresponds to the extracellular domain (aa 65-265) of human CLEC12A, which contains the C-type lectin domain and is sufficient for generating antibodies that recognize the native receptor on live cells.
- This conjugate is optimized for direct flow cytometry (FC) without a secondary antibody; consider titrating the antibody to determine the optimal working concentration for your cell type and staining conditions.
- For surface staining of CLEC12A, harvest cells and wash with FACS buffer (PBS containing 1–2% FBS or BSA). Incubate with the fluorophore-conjugated antibody at 2–8°C for 20–30 minutes, protected from light.
- Include appropriate isotype controls and Fc receptor blocking reagents to minimize non-specific binding, especially in myeloid cell populations.
- Data acquisition should be performed as soon as possible after staining; if delay is unavoidable, fix cells with paraformaldehyde after washing and protect from light.
- Avoid repeated freeze-thaw cycles of the antibody; store the conjugate undiluted at 2–8°C and protect from prolonged light exposure.
