Biological Background: CEACAM6 Function and Localization

  • CEACAM6 (CD66c) is a GPI-anchored cell surface glycoprotein that mediates cell adhesion in a calcium- and fibronectin-independent manner, playing a key role in tumor progression (PubMed:10910050, PubMed:11590190, PubMed:16204051).
    Related references: PMID:10910050 PMID:11590190 PMID:16204051
  • It mediates homophilic binding as well as heterophilic interactions with other CEACAM family members, including CEACAM5 and CEACAM8; the interaction with CEACAM8 occurs on activated neutrophils (PubMed:8776764, PubMed:11590190).
    Related references: PMID:8776764 PMID:11590190
  • CEACAM6 is localized primarily at the cell membrane, with apical enrichment on epithelial cells.
  • The protein is expressed in neutrophils and in columnar epithelial and goblet cells of the colon, and is frequently overexpressed in tumor cell lines.
  • Functional studies show that CEACAM6 promotes neutrophil adhesion to cytokine-activated endothelial cells and facilitates cell migration (PubMed:1378450, PubMed:16204051).
    Related references: PMID:1378450 PMID:16204051
  • As a member of the immunoglobulin superfamily, CEACAM6 contains multiple Ig-like domains and is modified by disulfide bonds and glycosylation; it is linked to the membrane via a GPI anchor.
  • Aberrant expression of CEACAM6 is associated with oncogenesis and apoptosis, making it a potential biomarker and therapeutic target.

Experimental Guidance and Technical Tips

  • The immunogen (aa 35-320) corresponds to the extracellular domain of CEACAM6; therefore, the antibody is predicted to recognize the native surface protein, making it well-suited for live-cell flow cytometry applications.
  • For flow cytometry, consider titrating the antibody to determine optimal staining concentration, and include appropriate isotype controls and single-stain controls for compensation.
  • Validate antibody performance in relevant human cell lines or primary cells known to express CEACAM6 (e.g., neutrophils, colon epithelial cells, or tumor cell lines). Fixation and permeabilization may affect staining due to the GPI anchor; assess compatibility experimentally.
