Biological Background: CEACAM5 Function and Localization
- CEACAM5 (Carcinoembryonic antigen-related cell adhesion molecule 5), also known as CEA or CD66e, is a cell surface glycoprotein and a member of the carcinoembryonic antigen (CEA) family encoded by the CEACAM5 gene (GeneID: 1048).
- It functions in cell adhesion, mediating both homophilic binding and heterophilic interactions with other CEACAM family members such as CEACAM6, and contributes to intracellular signaling pathways that influence tumor progression. Related references: PMID:10864933 PMID:10910050 PMID:2803308
- Plays an oncogenic role by promoting tumor progression and inducing resistance to anoikis (detachment-induced apoptosis) in colorectal carcinoma cells, enhancing metastatic potential. Related references: PMID:10910050
- As a receptor for E.coli Dr adhesins, CEACAM5 binding facilitates bacterial attachment and may contribute to infectious diarrhea pathogenesis.
- It is localized primarily at the apical cell membrane and cell surface, consistent with its role in cell adhesion and signal transduction.
- The protein is extensively glycosylated and attached to the membrane via a glycosylphosphatidylinositol (GPI) anchor; it contains immunoglobulin-like domains and disulfide bonds critical for its structure.
- Expression is predominantly in columnar epithelial and goblet cells of the colon, and it is highly upregulated in adenocarcinomas of the digestive system and fetal colon, making it a well-known tumor marker. Related references: PMID:10436421
Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 35-685 of human CEACAM5, which encompasses the entire extracellular domain. Thus, the antibody is likely to detect the native, membrane-bound conformation in flow cytometry assays.
- For flow cytometry, use an appropriate isotype control antibody (rabbit IgG, same fluorophore) to assess non-specific binding.
- Perform antibody titration (e.g., 0.1–10 μg/mL) on your cell line of interest to determine the optimal staining index.
- Choose a cell line with confirmed CEACAM5 expression (e.g., colorectal adenocarcinoma cells like HT-29 or LS174T) as a positive control, and a CEACAM5-negative line as a negative control.
- is a bright, photostable Alexa Fluor® 488 equivalent, allowing flexibility in multicolor panel design. Ensure compensation when using other green-channel fluorophores.
- For intracellular staining, consider a fixation and permeabilization protocol optimized for GPI-anchored cell surface proteins, as harsh permeabilization may disrupt membrane localization.
