Biological Background: CD94 Function and Localization
- CD94 (KLRD1) is a C-type lectin-like receptor that serves as the ligand-binding subunit of the CD94/NKG2 heterodimeric receptor complexes on natural killer (NK) cells and subsets of T cells. It lacks intrinsic signaling capacity, relying on its partner chains KLRC1 (NKG2A) or KLRC2 (NKG2C) for signal transduction.
- The CD94/NKG2A complex functions as an inhibitory receptor, recognizing the non-classical MHC class Ib molecule HLA-E bound to self-peptides derived from classical MHC class I signal sequences. This interaction enables NK cells to monitor MHC class I expression and maintain self-tolerance. Related references: PMID:10023772, PMID:18064301, PMID:18083576
- The CD94/NKG2C complex acts as an activating receptor, recognizing HLA-E loaded with peptides from HLA-G, and plays roles in adaptive NK cell responses and maternal-fetal tolerance. Related references: PMID:15940674, PMID:9655483, PMID:30134159, PMID:9754572, PMID:20952657, PMID:21825173
- CD94 is expressed on the cell membrane of NK cells, memory/effector CD8+ αβ T cells, and terminally differentiated γδ T cells, with differential expression of NKG2A and NKG2C in these populations.
- As a glycoprotein with a transmembrane domain and disulfide bonds, CD94 is classified as a receptor with innate and adaptive immunity roles, and it is involved in host-virus interactions.
- In viral infections, CD94/NKG2A can mediate immune evasion. During SARS-CoV-2 infection, CD94 may recognize HLA-E presenting viral spike protein peptides on lung epithelial cells, leading to NK cell exhaustion and reduced antiviral surveillance. Related references: PMID:32859121
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular region of CD94, suggesting the antibody binds a surface-exposed epitope.
- For flow cytometry, consider using human peripheral blood mononuclear cells or NK cell lines, and validate binding in the relevant sample system.
- Ensure optimization of antibody titration and appropriate isotype controls.
