Biological Background: CD93 Function and Localization
- CD93, also known as Complement component C1q receptor (C1qR, C1qRp), is a cell surface receptor encoded by the CD93 gene. It is a transmembrane glycoprotein belonging to the Group XIV C-type lectin family. Related references: PMID:7977768
- It functions as a receptor for defense collagens, including surfactant protein A (SFTPA1), complement component C1q, and mannose-binding lectin (MBL2), thereby enhancing phagocytosis of apoptotic cells and immune complexes. Related references: PMID:7977768
- CD93 plays a critical role in angiogenesis by regulating endothelial cell adhesion and migration. It associates with beta-dystroglycan to activate SRC kinase, leading to CBL recruitment and CRKL-mediated signaling. Related references: PMID:24809468 PMID:26848865
- It also serves as a receptor for pan-endothelial glycoprotein multimerin-2 (MMRN2) and IGFBP7, both of which contribute to CD93-mediated endothelial functions. Related references: PMID:28671670 PMID:36265539 PMID:38218180
- CD93 is highly expressed in endothelial cells, platelets, and myeloid cells (monocytes, neutrophils), but not in lymphoid cells, indicating a role in innate immunity and vascular biology.
- The protein undergoes post-translational modifications including phosphorylation, glycosylation, and disulfide bond formation, essential for its stability and function. It contains EGF-like domains.
- Keywords: Cell adhesion, Membrane receptor, Lectin, Host-virus interaction, Angiogenesis, Inflammatory response.
Experimental Guidance and Technical Tips
- This -conjugated antibody is optimized for direct flow cytometry detection of CD93 on live, unfixed cells. Since CD93 is a cell surface receptor, avoid permeabilization steps unless intracellular staining is required.
- Validate antibody performance in your specific cell type of interest. Positive controls include human endothelial cells, monocytes, or neutrophils, where CD93 is highly expressed; lymphoid cells serve as negative controls.
- Use an appropriate isotype control conjugated to the same fluorophore to assess nonspecific binding. Titrate the antibody to determine the optimal concentration for your assay.
- For dual-color flow cytometry, ensure spectral compatibility of with other fluorophores used. Note that has excitation/emission similar to Alexa Fluor 594.
- The immunogen spans amino acids 24-580, covering nearly the entire extracellular domain, which may enhance recognition of native CD93.
