Biological Background: CD9 Function and Localization

  • CD9, also designated as MIC3, MRP-1, BTCC-1, DRAP-27, and TSPAN29, belongs to the tetraspanin superfamily of integral membrane proteins.
  • It mediates sperm-egg fusion, platelet activation and aggregation, and regulates cell adhesion and motility, thereby influencing tumor metastasis. Related references: PMID:14575715, PMID:18541721, PMID:8478605, PMID:7511626
  • CD9 associates with integrins and other tetraspanins such as CD81 to modulate cell fusion processes, including macrophage multinucleation and myotube formation. Related references: PMID:12796480
  • The protein localizes to the plasma membrane and is also found in secreted extracellular exosomes.
  • Post-translational modifications include N-linked glycosylation and palmitoylation, with disulfide bonds contributing to structural stability.
  • CD9 is predominantly detected in platelets and broadly expressed in hematopoietic and epithelial cells. Related references: PMID:19640571

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 1–228 of human CD9, encompassing the large extracellular domain, which is the primary antigenic region for antibody recognition.
  • Direct conjugation to eliminates the need for secondary antibodies in flow cytometry, reducing background and protocol steps.
  • Consider titrating the antibody and including Fc receptor blocking in samples rich in macrophages or granulocytes.
  • Validate staining with relevant positive controls such as platelets, peripheral blood mononuclear cells, or CD9-expressing cell lines.
