Biological Background: CD87/PLAUR Function and Localization

  • Urokinase plasminogen activator surface receptor (uPAR, CD87) is a GPI-anchored glycoprotein encoded by the PLAUR gene, expressed on various cell types including immune cells and tumor cells.
  • Acts as a high-affinity receptor for urokinase-type plasminogen activator (uPA), leading to localized plasmin generation and subsequent extracellular matrix (ECM) degradation, facilitating cell migration and tissue remodeling. Related references: PMID:1689240 PMID:15677461
  • Independently binds the ECM protein vitronectin, promoting cell adhesion and indirectly activating integrin signaling pathways, which coordinate cell proliferation and survival. Related references: PMID:17548516 PMID:25168639 PMID:28849762
  • Found at the cell membrane, particularly enriched at invadopodia—actin-rich membrane protrusions mediating ECM degradation; a soluble secreted form is also generated by cleavage of the GPI anchor.

Experimental Guidance and Technical Tips

  • For flow cytometry, use live unfixed cells to preserve surface epitope integrity; if intracellular detection is required, fix/permeabilize with appropriate buffers.
  • The immunogen (aa 23–303) covers the entire extracellular domain. Gentle cell harvesting (e.g., non-enzymatic dissociation) is recommended to avoid cleavage of the GPI anchor and loss of surface staining.
  • Titrate the antibody to determine the optimal concentration for your specific cell type and experimental conditions, and include appropriate isotype controls.
  • As a direct conjugate, no secondary antibody is needed. Consider using a viability dye for dead cell exclusion.
