Biological Background: CD85j/ILT2 Function and Localization
- CD85j/ILT2, encoded by LILRB1 (also known as ILT2, LIR1, MIR7), is a leukocyte immunoglobulin-like receptor subfamily B member 1. It is a single-pass type I membrane protein with multiple immunoglobulin-like domains.
- Functions as an inhibitory receptor for a broad spectrum of MHC class I antigens, including HLA-A, HLA-B, HLA-C, HLA-G, and HLA-F, as well as the human cytomegalovirus class I MHC homolog UL18. Related references: PMID:16455647, PMID:28636952
- Ligand binding transmits inhibitory signals that down-regulate immune responses, such as inhibiting FCER1A-mediated serotonin release and FCGR1A-mediated phosphorylation and calcium mobilization. Related references: PMID:11907092, PMID:9285411, PMID:9842885
- Interaction with HLA-G in complex with B2M and self-peptide triggers secretion of growth-promoting factors by decidual NK cells and reprograms B cells toward an immune suppressive phenotype. Related references: PMID:19304799, PMID:29262349, PMID:24453251
- Subcellular localization includes the cell membrane and also secreted forms due to alternative splicing.
- The protein undergoes post-translational modifications such as phosphorylation and glycosylation, and contains disulfide bonds and immunoglobulin-like domains.
- Expressed in B cells, monocytes, dendritic cell subsets (myeloid, plasmacytoid, tolerogenic), decidual macrophages, and decidual NK cells.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 24–458), making this antibody suitable for detecting native surface CD85j/ILT2 in flow cytometry.
- Multiple protein isoforms exist (predicted weights 49 kDa, 70 kDa, 71 kDa); consider validating specificity in the relevant cell type under your experimental conditions.
- For optimal staining, titrate the antibody and include appropriate isotype controls, especially when analyzing cells with variable Fc receptor expression.
