Biological Background: LILRB2/CD85d/ILT4 Function and Localization
- Leukocyte immunoglobulin-like receptor subfamily B member 2 (LILRB2), also designated CD85d, ILT4, LIR2, and MIR10.
- Functions as a receptor for a broad spectrum of class I MHC antigens, including HLA-A, HLA-B, HLA-C, HLA-G and HLA-F alleles. Related references: PMID:11169396 PMID:12853576 PMID:16455647
- Involved in immune tolerance; recognizes HLA-G in complex with B2M and nonamer self-peptide to trigger differentiation of type 1 regulatory T cells and myeloid-derived suppressor cells, maintaining maternal-fetal tolerance. Related references: PMID:16455647 PMID:20448110 PMID:27859042
- Competes with CD8A for binding to class I MHC antigens and inhibits FCGR1A-mediated phosphorylation and intracellular calcium mobilization. Related references: PMID:11875462 PMID:12853576 PMID:9548455
- Localizes to the cell membrane.
- Expressed in monocytes, dendritic cells, NK cells, and B cells; notably enriched in tolerogenic IL10-producing dendritic cells and myeloid-derived suppressor cells during pregnancy. Related references: PMID:20448110 PMID:27859042
- Contains immunoglobulin domains, undergoes glycosylation and phosphorylation, and is subject to alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 22–461), providing targeting capability for cell surface-expressed LILRB2, suitable for flow cytometry applications.
- For flow cytometry, consider performing antibody titration to determine the optimal working concentration and include appropriate isotype controls to assess nonspecific binding.
- Validate the antibody in the relevant human sample system (e.g., peripheral blood mononuclear cells, dendritic cell subsets) to confirm specificity and sensitivity.
- Note that is a bright and photostable fluorophore; use standard blue laser (488 nm) excitation and 530/30 nm bandpass filter detection.
