Biological Background: CD84 Function and Localization

  • CD84, also known as SLAMF5, LY9B, or MAX.3, is a single-pass type I membrane protein belonging to the signaling lymphocytic activation molecule (SLAM) family.
  • As a self-ligand receptor, CD84 mediates homo- or heterotypic cell-cell interactions that modulate the activation and differentiation of innate and adaptive immune cells, with activities controlled by cytoplasmic adapters SH2D1A/SAP and SH2D1B/EAT-2.
  • Homophilic CD84 interactions enhance interferon gamma secretion in lymphocytes and induce platelet stimulation in a SH2D1A-dependent manner; CD84 may also serve as a marker for hematopoietic progenitor cells. Related references: PMID:11564780 PMID:12115647 PMID:12928397
  • In mast cells, CD84 negatively regulates high-affinity immunoglobulin epsilon receptor signaling, involving FES and PTPN6/SHP-1. Related references: PMID:22068234
  • CD84 positively regulates macroautophagy in primary dendritic cells by stabilizing IRF8 and inhibiting its TRIM21-mediated proteasomal degradation. Related references: PMID:29434592
  • CD84 is required for prolonged T-cell:B-cell contact, optimal T follicular helper function, and germinal center formation, and contributes to maintaining B-cell tolerance and preventing autoimmunity.
  • Subcellularly, CD84 localizes to the cell membrane and is predominantly expressed in hematopoietic tissues (lymph node, spleen, peripheral leukocytes) and cell types including macrophages, B-cells, monocytes, platelets, thymocytes, T-cells, dendritic cells, and mast cells.
  • Key post-translational modifications and structural features include a signal peptide, three immunoglobulin-like domains, disulfide bonds, glycosylation, phosphorylation, and alternative splicing, resulting in multiple protein isoforms.

Experimental Guidance and Technical Tips

  • The immunogen covers the extracellular domain (aa 22–225) of human CD84, targeting the native conformation. When designing flow cytometry panels, consider that CD84 is expressed on various immune cells; appropriate controls and gating strategies are recommended.
  • This conjugate is intended for flow cytometry; optimal dilution should be determined empirically by titration in the target sample system.
  • CD84 exists as multiple isoforms (observed molecular weight range 24–38 kDa); ensure that the detection system and sample preparation account for potential variability.
  • For functional studies, note that CD84 signaling is modulated by adapters such as SAP and EAT-2, so interpretation may depend on the cell type and activation state.
