Biological Background: CD84 Function and Localization
- CD84 (SLAM family member 5, LY9B, SLAMF5) is a single-pass type I membrane protein and a self-ligand receptor of the signaling lymphocytic activation molecule (SLAM) family.
- SLAM receptors triggered by homo- or heterotypic cell-cell interactions modulate the activation and differentiation of a wide variety of immune cells, bridging innate and adaptive immunity.
- CD84 enhances proliferative responses of activated T-cells, promotes interferon-gamma secretion in lymphocytes, and induces platelet activation via SH2D1A-dependent pathways.
- CD84 may serve as a marker for hematopoietic progenitor cells. Related references: PMID:11564780, PMID:12115647, PMID:12928397
- CD84 is required for prolonged T-cell:B-cell contact, optimal T follicular helper function, and germinal center formation, contributing to B-cell tolerance maintenance.
- In mast cells, CD84 negatively regulates high affinity immunoglobulin epsilon receptor signaling independent of SH2D1A and SH2D1B, involving FES and PTPN6/SHP-1. Related reference: PMID:22068234
- In macrophages, CD84 enhances LPS-induced MAPK phosphorylation and NF-kappaB activation, modulating cytokine secretion via ITSM 2.
- CD84 positively regulates macroautophagy in primary dendritic cells by stabilizing IRF8 and inhibiting TRIM21-mediated proteasomal degradation. Related reference: PMID:29434592
- Predominantly expressed in hematopoietic tissues (lymph node, spleen, peripheral leukocytes) and immune cell subsets including macrophages, B-cells, monocytes, platelets, thymocytes, T-cells, dendritic cells, and mast cells, with high expression in memory T-cells.
Experimental Guidance and Technical Tips
- For flow cytometry, titrate the antibody to determine the optimal concentration for your specific cell type and experimental conditions.
- Use an appropriate isotype control and ensure single-cell suspension to minimize nonspecific binding.
- Protect the fluorochrome-conjugated antibody from light during storage and handling; avoid repeated freeze-thaw cycles.
- Validate the antibody performance in your sample system, considering cell viability and potential Fc receptor blocking if needed.
