Biological Background: CD84 Function and Localization

  • CD84 (SLAM family member 5, LY9B, SLAMF5) is a single-pass type I membrane protein and a self-ligand receptor of the signaling lymphocytic activation molecule (SLAM) family.
  • SLAM receptors triggered by homo- or heterotypic cell-cell interactions modulate the activation and differentiation of a wide variety of immune cells, bridging innate and adaptive immunity.
  • CD84 enhances proliferative responses of activated T-cells, promotes interferon-gamma secretion in lymphocytes, and induces platelet activation via SH2D1A-dependent pathways.

  • CD84 is required for prolonged T-cell:B-cell contact, optimal T follicular helper function, and germinal center formation, contributing to B-cell tolerance maintenance.
