Biological Background: CD82 Function and Localization
- CD82, also known as KAI1, is a member of the tetraspanin family (TSPAN27) that functions as a metastasis suppressor.
- It is a structural component of tetraspanin-enriched microdomains (TERMs) that facilitate receptor clustering and signaling. Related references: PMID:19497983
- CD82 attenuates EGF signaling by promoting ligand-induced endocytosis and desensitization of EGFR, mediated by CBL phosphorylation via PKC. Related references: PMID:10985391 PMID:35538033 PMID:23897813
- It enhances cell-matrix adhesion by regulating integrin α4 and suppresses cell migration through integrins α6 and β1. Related references: PMID:24623721 PMID:8757325 PMID:15557282 PMID:17560548
- CD82 interferes with urokinase-type plasminogen activator (PLAU) binding to its receptor PLAUR, thereby reducing plasminogen activation. Related references: PMID:15677461
- It delivers costimulatory signals for the TCR/CD3 pathway through association with CD4 or CD8.
- CD82 inhibits LPS-induced inflammatory response by preventing LPS binding to TLR4, and contributes to TLR9 trafficking and myddosome assembly. Related references: PMID:36945827
- It inhibits angiogenesis by binding and sequestering pro-angiogenic factors VEGFA and PDGFB, switching the milieu to a quiescent state. Related references: PMID:34530889
- Subcellularly, CD82 localizes to the cell membrane, cytoplasmic vesicles, and phagosomes; it is post-translationally modified by palmitoylation and glycosylation.
Experimental Guidance and Technical Tips
- For flow cytometry, titrate the antibody to determine the optimal working concentration for your specific cell type and experimental conditions.
- Use appropriate isotype controls and viability dyes to ensure accurate gating and exclusion of non-specific binding.
- The immunogen corresponds to the full-length human CD82 protein; while the antibody is likely to recognize native epitopes, confirm performance with relevant positive and negative cell lines in your assay.
- Since CD82 is expressed on lymphoid cells, consider using peripheral blood mononuclear cells (PBMCs) or lymphoid cell lines as positive controls.
- The fluorophore is compatible with standard 488 nm laser lines; follow standard protocols for sample preparation and acquisition.
