Biological Background: CD80/B7-1 Function and Localization
-
CD80, also known as B7-1, is a T‑lymphocyte activation antigen and a costimulatory molecule belonging to the immunoglobulin superfamily that is essential for T‑cell priming. It provides the primary auxiliary signal augmenting the MHC/TCR signal in naive T‑cells by binding to the CD28 receptor.

CD80 engagement of CD28 activates downstream signaling pathways, including NF‑κB and MAPK cascades, leading to the production of cytokines such as IL‑2.

PMID:10438913 -
CD80 can also function as an inhibitor of T‑cell activation by acting as a ligand for CTLA4, a decoy receptor that outcompetes CD28 and blocks T‑cell priming.

The CD28/CD80 costimulatory signal promotes T‑cell glucose metabolism and ATP synthesis through activation of the PI3K/Akt pathway.

CD80 regulates PD‑L1/PD‑1 interactions, limiting excessive PD‑L1 engagement and its inhibitory impact on immune responses.

In B‑cells, CD80 is critical for regulating B‑cell/T‑cell interactions during early and late germinal center responses, which are necessary for effective humoral immunity. (By similarity)
-
CD80 localizes to the cell membrane and is expressed on the surface of antigen‑presenting cells (APCs), including dendritic cells, macrophages, and activated B‑cells.
-
The protein undergoes alternative splicing and post‑translational modifications such as glycosylation and palmitoylation. It also serves as a receptor for adenovirus subgroup B during microbial infection.
Experimental Guidance and Technical Tips
- This ‑conjugated rabbit monoclonal antibody is designed for flow cytometry (FC). Perform titration experiments to determine the optimal working concentration for your specific sample type.
- CD80 is expressed on the surface of antigen‑presenting cells; use relevant human cell lines (e.g., Raji, THP‑1) or primary cells (e.g., peripheral blood mononuclear cells, dendritic cells) as positive controls.
- is a far‑red fluorophore excitable at ~650 nm with emission near 665 nm. Ensure proper compensation, especially in multicolor panels, and avoid fluorophore combinations with significant spectral overlap.
- Include appropriate isotype controls and viability dyes to exclude dead cells and minimize nonspecific binding.
- For intracellular staining (e.g., after Brefeldin A treatment), fix and permeabilize cells before antibody incubation, and verify that the epitope remains intact.
- The immunogen corresponds to the extracellular domain (aa 35‑242); thus the antibody is suitable for staining of live, non‑permeabilized cells. Avoid harsh fixation that may alter epitope recognition.
