Biological Background: CD7 Function and Localization

  • CD7 is a transmembrane glycoprotein belonging to the immunoglobulin superfamily, also known as GP40, LEU-9, Tp40, and TP41. It contains an immunoglobulin domain and is modified by disulfide bonds, glycosylation, and palmitoylation.
  • CD7 functions as a costimulatory receptor on T-cells and NK cells, facilitating full activation upon binding to its ligand K12/SECTM1. Related references: PMID:10652336
  • In T-cells, CD7 engagement promotes the production of interleukin-2 (IL-2), a critical growth factor for T-cell proliferation. Related references: PMID:1709867
  • On resting NK cells, CD7 activation induces a significant increase in interferon-gamma (IFN-γ) secretion, enhancing innate immune responses. Related references: PMID:7506726
  • CD7 is specifically expressed on the surface of T-lymphocytes and NK cells, as well as their precursors, consistent with its role in adaptive and innate immunity.
  • The protein is localized to the plasma membrane, with a single-pass transmembrane helix and a cytoplasmic tail that contains palmitoylation sites, anchoring it to membrane lipid rafts.

Experimental Guidance and Technical Tips

  • For flow cytometry, titrate the antibody to determine the optimal staining concentration for your cell population, and always include appropriate isotype controls.
  • Since CD7 is expressed on T and NK cells, validate staining on peripheral blood mononuclear cells (PBMCs) or isolated lymphocyte subsets.
  • Consider fixation and permeabilization protocols if intracellular staining is required, ensuring compatibility with the fluorophore.
