Biological Background: CD7 Function and Localization
- CD7 is a transmembrane glycoprotein belonging to the immunoglobulin superfamily, also known as GP40, TP41, and Leu-9, encoded by the CD7 gene.
- It is expressed on T-cells, natural killer (NK) cells, and their precursors.
- CD7 functions as a costimulatory receptor; upon binding to its ligand K12/SECTM1, it enhances T-cell activation and promotes IL-2 cytokine production. Related references: PMID:10652336 PMID:1709867
- On resting NK cells, CD7 engagement leads to significant induction of interferon-gamma levels. Related references: PMID:7506726
- The protein is localized at the cell membrane as a single-pass type I membrane protein.
- Post-translational modifications include disulfide bond formation, glycosylation, and palmitoylation, which may regulate its trafficking and function.
- Keywords: Adaptive immunity, Immunity, Immunoglobulin domain, Receptor, Transmembrane helix.
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fusion protein covering amino acids 26-180 of human CD7 (NP_006128.1), comprising the extracellular domain; thus, the antibody likely recognizes an extracellular epitope, suitable for live-cell flow cytometry.
- For flow cytometry (FC), titrate the antibody to determine optimal concentration for your specific cell type and experimental conditions; a typical starting concentration may be 0.5–5 µg/mL, but validation is recommended.
- Protect the antibody from light, as it is conjugated to the photosensitive fluorophore, and store at 2–8°C upon receipt; avoid repeated freeze-thaw cycles.
- Cross-reactivity has been validated for human; verify reactivity with other species if needed.
- The antibody is supplied in a liquid format with ice bag shipping; ensure proper cold chain maintenance.
